No, she'd have been Queen no matter who she married. It could've provoked a civil war (hence why she wasn't really keen on marrying an Englishman) but the act of marriage in and of itslef would in no way affect her legal claim to the throne or her status as Queen Regnant.
Unless he was Catholic. She couldn't have married him if he was Catholic.
